What the corpus says

Pinealgland polypeptide fraction. Khavinson's 1970s predecessor to the defined tetrapeptide Epitalon. Largely replaced in clinical use once the shorter, synthesizable sequence was characterized.

Mechanism

Polypeptide fraction from bovine pineal gland (epiphysis). the parent fraction from which AEDG (epitalon) was derived. The mechanism is essentially the pineal-gland-substitute role: epithalamin increases lifespan in various animal species and inhibits spontaneous carcinogenesis (Grigoriev-Korneenkov-Popovich 2019 statistical methodology paper for the clinical trial of epithalamin). Pineal peptides including the AEDG tetrapeptide regulate neuro-immuno-endocrine balance. The Russian clinical literature (Grigoriev 2019) on epithalamin explicitly raises statistical methodology issues. Bonferroni correction, Bayes factors, p<0.005 vs p<0.05. because the trials are Russian-only and small. Epithalamin has direct reproductive endocrine effects (Galnykina 2008: pineal regulation of testicular endocrine tissue), and is part of the broader pineal-reproductive axis (Pishak 2012).

Key findings
  1. Pinealectomised rats: epithalamin restores spleen + thyroid balance after pinealectomy (Linkova 2011, same paper as epitalon)
  2. Animal lifespan: epithalamin increases lifespan in various species and inhibits spontaneous carcinogenesis. basis for clinical study in elderly age-related pathology prevention (Grigoriev 2019)
  3. Pineal regulation of testicular endocrine tissue (Galnykina 2008). direct reproductive axis effect
  4. Review of pineal role in male reproductive system (Pishak 2012). broader pineal-reproductive axis
  5. Statistical methodology critique: Grigoriev 2019 notes that long-term epithalamin clinical studies in elderly raise multiple-comparisons issues that classical p<0.05 analyses may have over-interpreted
Evidence quality

grade: Moderate. Strong in animal models, weaker in clinical translation (Russian-only trials, small n, statistical methodology concerns explicitly raised by Russian authors themselves).

Open questions
  • Is the lifespan extension in animals reproducible by independent labs?
  • Pineal-reproductive axis: is the mechanism melatonin-mediated or peptide-mediated or both?
  • Clinical efficacy in elderly age-related pathology. meta-analysis with corrected statistics
  • Active principle: which peptides within the pineal fraction drive the effect (vs the AEDG tetrapeptide alone)?
